A white crystalline solid, 1-phenethylbiguanide hydrochloride, C10H15N5.HCl, which is used in the oral treatment of diabetes. Also phenformin hydrochloride.

LXXX. 773/1 In 1957 Ungar, Freedman and Shapiro reported that hypoglycæmic properties had been discovered in a synthetic diguanide compound, N′-β-phenethylformaminyliminourea hydrochloride, subsequently designated DB1 or phenformin. 1960[see biguanide]. 1961Lancet 9 Sept. 566/1 Phenformin is clearly a potent hypoglycæmic substance effective in most diabetics we have tested. 1967Martindale's Extra Pharmacopoeia (ed. 25) 675/2 Phenformin hydrochloride is contra-indicated in acidosis, coma, infections, [etc.]. 1974M. C. Gerald Pharmacol. xxv. 441 It appears that phenformin may act by accelerating the intracellular oxidation of glucose, a process which is depressed in the absence of insulin. Phenformin is used alone for the treatment of maturity-onset diabetes and in combination with insulin in juvenile diabetes. 1977Lancet 22 Jan. 191/2 Metformin is replacing phenformin as the biguanide of choice in the treatment of obese maturity-onset diabetics because of the association between phenformin therapy and lacticacidosis. |
